b&g$y, With the aim of tmproving early dtiagnosis In infectious weitts, the results of PCR and serological anel~sis of intraocular fluads wwe evaluated. b$&~&. Aqueous humor or vitmous samples of panrntr with dcula re~mal nccros~s IARN;” z. 161, rotm~trs 4” AlDS (n 381, toxoplasma choriorerhwis cn = 221 and different uvorfis entmcs (n 371 were te:itad lor the presence of Varicalla-zoster wus, Herpes simplex virus, Cbromogalovirus and TOXODlaSma DNA ~4th PCR. The same sampler w:ro alS0 tested for local antibody produclion by calculating tha Gc ldmann Witmor coofftcionr aBains1 the aforemontionod agents. f&nrclu_ In ARN, the GW-coefficient for VZV or HSV was positive in 6S% and PCR for VZV or ~ISV DNA in 50%. Combination of both techniques yielded a posttlvo lost result In 81%. In toxoplasma ch0riore!init& 68% had a posrtiva GW-coefficient for toxoplasms and PCH was positive in 18%. None of the ocular fluid samples of ,mmunocompet0nt persons was positwe for CMV. In patients with AIDS. PCR was poslrivo for CMV DNA In 61% of the OCdar fluia sanples tested. Four ocular flwd samples of AIDS patients w0re positive for VZV DNA and thr00 of lheae patients ware diagnosed as progressive outfr retinal necrosis. The GW-coelhci0nt was negative I” 311 the patients wrrh AIDS dospirc the prescncr? ot harpesviral DNA. Ccntrol aqueous or virroous samples In =38) did not contain dotoctable horposwrsl or toxoplasma DNA. J;PIJ&~ The combination ot PCA and local antibody ptoductior, Improvas the datcctiqn of infectious causes II-I uveius in immunocwnpetont parsons In patients with AIDS, PCR is a more swsitivo techniqua to establish on etiologic dlagnosls than ,wwlysra 01 local antibody pruduction.

Novel multi-targeted polymerase chain reaction for diagnosis of presumed tubercular uveitis

BACKGROUND The objective of this study was to report the use of multi-targeted polymerase chain reaction (PCR) in the diagnosis of presumed tubercular uveitis. Multi-targeted PCR using three targets specific for Mycobacterium tuberculosis, i.e., IS6110, MPB64, and protein b, was performed on intraocular fluid samples of 25 subjects. Nine had presumed tubercular uveitis, six had intraocular infl...

Increased presence of Epstein-Barr virus DNA in ocular fluid samples from HIV negative immunocompromised patients with uveitis.

AIMS To investigate whether routine testing for Epstein-Barr virus (EBV) is necessary in the examination of a patient with uveitis. METHODS Intraocular EBV DNA was determined in 183 ocular fluid samples taken from patients with AIDS and uveitis, HIV negative immunocompromised uveitis, acute retinal necrosis, toxoplasma chorioretinitis, intraocular lymphoma, anterior uveitis, and miscellaneous...

Diagnosis of ocular toxocariasis by establishing intraocular antibody production.

PURPOSE To investigate the role of Toxocara canis in posterior uveitis of undetermined origin. DESIGN Retrospective case-study. METHODS Paired ocular fluid (47 aqueous humor [AH] and two vitreous fluids) and serum samples of 37 adults and 12 children with undetermined posterior uveitis were retrospectively analyzed for intraocular IgG antibody production against Toxocara canis by enzyme-lin...

The Value of Routine Polymerase Chain Reaction Analysis of Intraocular Fluid Specimens in the Diagnosis of Infectious Posterior Uveitis

OBJECTIVE To assess the value of routine polymerase chain reaction (PCR) analysis on intraocular fluid from patients presenting with a first episode of suspected active infectious posterior uveitis in a population with a high prevalence of human immunodeficiency virus infection. DESIGN Retrospective, interventional case series. Participants. 159 consecutive patients presenting at a tertiary c...

Diagnosis of bacterial endophthalmitis by broad-range quantitative PCR.

AIM To measure the bacterial genome in ocular fluids and to analyse the clinical relevance of infectious endophthalmitis. METHODS Nineteen ocular fluid samples (eight aqueous humour and 11 vitreous fluid samples) were collected from 19 patients with suspected bacterial endophthalmitis. Fifty ocular samples from uveitis patients were also collected along with 40 samples from patients without o...
